In order to identify algae species trough metabarcoding techniques, a representative environmental sampling technique and reproducible eDNA extraction method have to be established. Throughout this secondment, in various location in the south of Vietnam, environmental samples have been taken with different filter techniques. Furthermore, an eDNA extraction protocol was used in order to isolate different levels of eDNA amounts. Trough PCR and the well established rbcL marker for metabarcoding, the amplified marker region was generated and sent for further sequencing.
